Buclizine
Mechanism :
Buclizine is a piperazine antihistamine with antimuscarinic and moderate sedative properties. It is used mainly for its antiemetic action and in the treatment of migraine in combination with analgesics.
Indication :
- Prophylaxis of motion sickness
- Migraine
Contraindications :
Absolute: Hypersensitivity to Buclizine or any of its components, Neonates.
Relative: Glaucoma (angle closure or predisposition to it), gastrointestinal obstruction.
Dosing :
Migraine:
10-14 years: 6.25 mg at the start of the symptoms.
>14 years (same as adult dose): 12.5 mg at the start of the symptoms.
Motion sickness:
Adults: 25 or 50 mg given 30 min before travelling, repeated after 4-6 hours, if necessary.
Adverse Effect :
Drowsiness, dizziness, incoordination, blurred vision, urinary retention, vomiting, rash, dry mouth, headache, nausea, epigastric pain, diarrhea, weight gain.
Interaction :
CNS depressants/alcohol: Concurrent use with other CNS depressants/alcohol may potentiate the CNS depression effects of buclizine.
Anticholinergics: Concurrent use with other anticholinergics may potentiate the anticholinergic effects.
Apomorphine: Prior administration of buclizine with apomorphine decreases the emetic response to apomorphine.
Hepatic Dose :
No dosage adjustments are recommended. Use with caution in liver disease.
