Sarecycline
Mechanism :
It is a tetracyclines which inhibits protein synthesis by binding with the 30S and possibly the 50S ribosomal subunit(s) of susceptible bacteria.
Indication :
- Treatment of inflammatory lesions of non-nodular moderate to severe acne vulgaris in patients ≥9 years.
Contraindications :
Hypersensitivity to sarecycline, tetracyclines, or any component of the formulation.
Dosing :
Use in children ≥9 years of age.
33 to 54 kg:
60 mg PO once daily.
55 to 84 kg:
100 mg PO once daily.
85 to 136 kg:
150 mg PO once daily.
Adverse Effect :
Nausea, vulvovaginal infections.
Interaction :
Antacids: May decrease the absorption of Tetracyclines.
Calcium Salts: May decrease the serum concentration of Tetracyclines.
Digoxin: Sarecycline may increase the serum concentration of Digoxin.
Iron Salts: Tetracyclines may decrease the absorption of Iron Salts.
Neuromuscular-Blocking Agents: Tetracyclines may enhance the neuromuscular-blocking effect of Neuromuscular-Blocking Agents.
Hepatic Dose :
No clinical differences in the pharmacokinetics of sarecycline were noted in mild to moderate hepatic impairment; the effect of severe hepatic impairment has not been studied.
