Lisdexamfetamine
Mechanism :
The exact mechanism is not known. Lisdexamfetamine dimesylate is a prodrug that is converted to the active component dextroamphetamine
Indication :
- Attention-deficit/hyperactivity disorder
- Binge eating disorder
Contraindications :
Hypersensitivity to amphetamine products or any component of the formulation; concurrent use of MAO inhibitor, or within 14 days of the last MAO inhibitor dose.
Known hypersensitivity or idiosyncrasy to sympathomimetic amines; advanced arteriosclerosis; symptomatic cardiovascular disease; moderate-to-severe hypertension; hyperthyroidism; glaucoma; agitated states; history of drug abuse.
Dosing :
To be used in children ≥6 years and adolescents orally.
Initial: 30 mg once daily in the morning; may increase in increments of 10 mg or 20 mg at weekly intervals; maximum: 70 mg/day.
Adverse Effect :
Insomnia, xerostomia, upper abdominal pain, tachycardia, hypertension, palpitations, Irritability, anxiety, jitteriness, agitation, hyperhidrosis, skin rash, weight loss, vomiting, diarrhea, erectile dysfunction, tremor.
Interaction :
Antihypertensive Agents: Amphetamines may diminish the antihypertensive effect of Antihypertensive Agents.
Antipsychotic Agents: May diminish the stimulatory effect of Amphetamines.
Monoamine Oxidase Inhibitors: May enhance the hypertensive effect of Amphetamines.
Tricyclic Antidepressants: May enhance the stimulatory effect of Amphetamines. Tricyclic Antidepressants may also potentiate the cardiovascular effects of Amphetamines.
