Pefloxacin

Mechanism :

Pefloxacin is a fluoroquinolone antibacterial that exhibits its bactericidal action by inhibition of DNA gyrase and topoisomerase IV which are needed for bacterial DNA reproduction.


Indication :

  • Uncomplicated gonococcal urethritis in males
  • Gastrointestinal system and genitourinary tract infection caused by gram negative bacteria.

Contraindications :

Hypersensitivity to quinolones, children <12 years, pregnancy, lactation.


Dosing :

15-20 mg/kg per day PO bid for 14-28 days.

Adverse Effect :

Hives, swelling of the face, lips, tongue or throat and in severe cases difficulty in breathing, gastric pain, nausea, vomiting, photosensitivity, muscular pain and headache.


Interaction :

Antacids, Didanosine (DDI), Iron Salts, Pirenzepine, Sucralfate, Vincristine, Zinc Salts: Reduced absorption.

Cimetidine, Probenecid, Theophylline: Increased levels.

Foscarnet, NSAIDs, Piroxicam: Increases the levels of cyclosporine, causes seizures.

Glimepiride: Hypoglycemia.

Quercetin: Antagonistic.



Hepatic Dose :

There are no specific pediatric recommendations; based on experience in adult patients, dosing adjustment is suggested.
